一种水溶性党参多糖的分离纯化及结构分析

摘要:

采用水提醇沉法得到党参粗多糖(COP),采用Sevag法除去蛋白成份,接着通过Sephacry1 S-200HR及Sephadex G-25凝胶柱色谱分离得到均一多糖COP-1。凝胶渗透色谱法(HPGPC)测定其纯度和平均分子量。高效液相色谱法(HPLC)确定其单糖的组成。红外光谱及核磁推测COP-1结构。结果表明COP-1平均分子量约为2.1×103 Da,且均由阶D-(2→1)呋喃果糖组成。

Abstract:

Crude Codonopsis pilosula polysaccharide was gained using water extraction-alcohol precipitation,and then Sevag method was used to remove protein.The main fraction of Codonopsis pilosula polysaccharide,namely COP-1,was isolated by Sephacryl S- 200 HR and Sephadex G- 25 column chromatography.High Performance Gel Permeation Chromatography(HPGPC) was used to determinate the purity and molecular weight of COP-1.High Performance Liquid Chromatography(HPLC) was used to analyze the monosaccharide composition of COP-1.Its structure was elucidated by FT-IR and NMR(1H-NMR,13C-NMR,HMBC and HSQC).The molecular weight of COP-1 was about 2.1×103 Da,and the mainly constitution of it was β-D-(2→1) fructose.
