PIWI蛋白特异性地在动物生殖系细胞中表达,对于动物生殖细胞发育分化至关重要。PIWI蛋白可以特异性地与一类被称为PIWI相互作用RNA的小分子非编码RNA结合。它们组装成功能性的复合体从而在转录水平或者转录后水平抑制转座元件的活性来维持基因组的稳定性。本研究系统总结了关于PIWI蛋白功能,piRNA的产生机制及功能,以及PIWI/piRNA复合体及其他蛋白因子在小鼠精子发生过程中发挥功能的分子机制等方面的相关研究进展,这些发现有助于我们更好地了解PIWI/piRNA在精子发生中的功能机制,并为相关男性不育症的诊治提供了理论依据和方法技术。
Abstract:PIWI proteins are specifically expressed in animal germ cells, which are essential for germline development. PIWI-interacting RNAs(piRNAs) are a novel class of small noncoding RNAs that are associated with PIWI proteins specifically. The PIWI/piRNA complex can prevent the activity of transposable elements and protect genome integrity via transcriptional or posttranscriptional mechanisms in animal germ cells. This review summarized the recent progress of the function of PIWI proteins, the mechanism of piRNA biogenesis and functional molecular of PIWI/piRNA complex and other proteins in mice spermatogenesis. These findings brought new insight into the functions and mechanisms of PIWI/piRNA in mammalian spermatogenesis, providing theory and technology for diagnosis and treatment of human male infertility.
