砂藓MYB转录因子基因RcMYB的克隆及表达分析

摘要:

以砂藓(Racomitrium canescens)转录组测序获得的一条与抗旱相关的MYB转录因子基因为基础,采用RT-PCR技术克隆得到该基因的cDNA全长序列,命名为RcMYB。该序列全长为862bp,开放阅读框为726bp,共编码氨基酸241个。该基因的蛋白相对分子质量为28.8 kD,等电点为7.77,不稳定系数为70.03,是不稳定性蛋白,平均亲水数为-0.548,预测分析该蛋白具有强亲水性。蛋白结构域分析表明,RcMYB蛋白含有Myb-DNA结合结构域和MYB-CC结构域。实时荧光定量PCR研究表明,在复水过程和干旱处理的条件下RcMYB基因均有所表达。上述研究结果为基因RcMYB的功能研究奠定基础。

Abstract:

A full length c DNA sequence of MYB transcription factor gene was cloned from Racomitrium canescens using RT-PCR, named as Rc MYB. Rc MYB was 862 bp in length, and open reading frame of 726 bp,encoding a protein of 241 amino acids. Protein relative molecular mass of gene was 28.8 k D and p I was 7.77.Unstable coefficient is 70.03 and showed this protein was instabile, and average number of hydrophilic was-0.548. Prediction and analysis of sequence showed the protein was highly hydrophilic. Protein domain analysis showed that Rc MYB protein contained MYB DNA-binding domain and MYB-CC domain. The q RT-PCR results showed that Rc MYB gene could express in water recovery process and drought treatment. The results would lay the foundations for the study of Rc MYB gene function.
