经试验,初步总结出一个适于甘蔗测酶活性的系统组成。用0.1mol/LpH6.0磷酸缓冲液作反应介质,测到的抗坏血酸氧化酶活性比用水作反应介质提高2.7倍。DTT对抗坏血酸氧化酶活性有较强的抑制作用,测定抗坏血酸氧化酶活性时,底物(抗坏血酸)的适宜pH为5.0~6.0,测定多酚氧化酶活性时,底物(邻苯二酚)的pH应比抗坏血酸的pH高1.0单位为佳。
Abstract:A suitable reaction system for determining the activities of ascorbic acid oxidase and polyphenol oxidase in sugarcane(Saccharum spp.)leaf was established.It was found that the activity of ascorbic acid oxidase was 2.7 times higher when a 0.1 mol/L pH 6.0 phosphate buffer was used as the reaction medium compared with the distilled water medium.Ascorbic acid oxidase activity was strongly inhibited by DTT.The optical pH of ascorbic acid was found to range from 5.0 to 6.0 for determining the activity of ascorbic acid oxidase,while that of pyrocatechol should be one pH uuit higher than that of ascorbic acid when determining the activity of polyphenol oxidase.
