本研究旨在探讨摄入槲皮素搭配跑台干预对阿霉素抗肿瘤的效果增强作用和阿霉素心脏毒性的缓解作用,为开发减轻化疗后癌症患者毒副作用的辅助方案提供理论依据。本研究将60只SPF小鼠随机分配对照组(C组, n=15)、阿霉素组(D组, n=15),槲皮素组(不运动+阿霉素+槲皮素; QD, n=15)、运动组(运动+阿霉素;简称SD组, n=15)、槲皮素+运动组(运动+槲皮素+阿霉素; SQD, n=15)。SD组和SDQ组加入12周跑台运动训练,QD组和SDQ组摄入槲皮素。SD组、QD组和SDQ组在进行实验开始后第5天开始,每天从腹腔注射方式对小鼠进行DOX给药,连续15 d。C组小鼠在同一周期内每日注射等量的生理盐水。8周干预结束后,使用心脏超声波分析小鼠的心脏功能。一方面,QD组、SD组、SDQ组PWs高于D组(p<0.05);另一方面,SDQ组PWd高于D组(p<0.05)。D组MV小于C组,大于D组和SD组(p<0.05)。D组AV小于C组,大于QD组、D组和SD组(p<0.05)。SDQ组FS大于D组(p<0.05)。本研究认为槲皮素搭配运动对于对阿霉素诱发的心毒性(心脏形态变化和心脏功能变化)具有一定预防作用。这一结果为槲皮素作为新辅助化疗的联合用药提供了理论可能性。
Abstract：The purpose of this study was to investigate the effects of quercetin in combination with treadmill intervention on the antitumor effect of adriamycin and the alleviating effect of adriamycin on cardiotoxicity, and to provide a theoretical basis for the development of an adjuvant scheme to reduce the toxic side effects of cancer patients after chemotherapy. In this study, 60 SPF mice were randomly assigned to the control group(group C, n=15), the doxorubicin group(group D, n=15), and the quercetin group(non-exercise+doxorubicin+quercetin). QD,n=15), exercise group(exercise +doxorubicin; SD group for short, n=15), quercetin+exercise group(exercise +quercetin+doxorubicin; SQD, n=15). SD group and SDQ group were added treadmill training for 12 weeks, QD group and SDQ group received quercetin. The SD group, QD group, and SDQ group started on the 5 th day after the experiment was started, and the mice were administered DOX by intraperitoneal injection every day for 15 consecutive days. Mice in group C were injected with the same amount of normal saline daily in the same cycle.After the 8-week intervention, the heart function of the mice was analyzed using echocardiography. On the other hand, the PWs in QD group, SD group, and SDQ group were higher than those in D group(p<0.05). In terms of PWd, SDQ group was higher than D group(p<0.05). The MV of group D was smaller than that of group C, and larger than that of group D and SD(p<0.05). The AV in group D was smaller than that of group C and larger than that of QD, D and SD groups(p<0.05). The SDQ group had greater FS than the D group(p<0.05). This study believes that quercetin combined with exercise has a certain preventive effect on adriamycin-induced cardiac toxicity(changes in cardiac morphology and cardiac function). This result provides theoretical possibilities for quercetin as a combination for neoadjuvant chemotherapy.
