龙牙百合多糖对人肝癌细胞HepG2凋亡的影响

摘要:

为研究龙牙百合多糖(LP)对人肝癌HepG2细胞抑制和凋亡的影响,采用MTT法考察了3组LP不同浓度对人肝癌HepG2细胞抑制作用,采用Annexin-V/PI双染法和Western blotting法探究了LP1和LP42个组分对人肝癌HepG2细胞凋亡作用机制。LP对人肝癌HepG2细胞抑制呈剂量依赖关系,LP4组1 mg/mL对HepG2细胞的抑制率达46.36%,效果最佳;LP1组8 mg/mL最高抑制率达43.23%;LP2组4 mg/mL最高抑制率达24.35%;LP1和LP4对HepG2细胞的晚期凋亡率显著高于早期。激活Caspase-3、Bax、蛋白表达显著升高(p<0.05),Bcl-2蛋白表达水平明显降低(p<0.05)。研究表明,LP通过激活死亡受体凋亡途径和线粒体途径,诱导人肝癌HepG2细胞凋亡。

Abstract:

In order to study the effects of Lilium polysaccharides(LP) on the inhibition and apoptosis induced in HepG2 cells, MTT assay was used to perform the inhibitory effect on HepG2 cells at certain tme intervals after incubation of various concentrations of LP. The protein levels of apoptosis in HepG2 cells with LP1 and LP4 were detected by Western blotting. Our research shows that LP reduced viability and proliferation of the HepG2 cells dose-dependently to some extent. The best inhibition rate of HepG2 cells was 46.36% in LP4 group with 1 mg/mL,followed by the highest inhibition rate of 43.23% in LP1 group with 8 mg/mLand the highest inhibition rate of 24.3% in LP2 group with 4 mg/mL. The late apoptotic rate of LP1 and LP4 cells was significantly higher than that of early apoptotic cells. Up-regulated the ratio of caspase-3, Bax, and down-regulated the ratio of Bcl-2 after treatment with SP. Above study, LP induces apoptosis of human HepG2 cells by activating death receptor apoptosis pathway and mitochondrial pathway.
