三角梅下胚轴再生体系的建立

本研究以三角梅(Bougainvillea glabra)品系‘Bouform 1’种子下胚轴为外植体,通过研究在MS培养基上添加不同的植物生长调节剂,其在诱导分化以及不定芽增殖、壮苗、生根等阶段的生长状况,以期筛选出最适宜其生长的培养基,建立稳定有效的再生体系。结果表明:下胚轴诱导分化最佳培养基为MS+IBA(吲哚丁酸)0.1 mg/L+6-BA(6-苄氨基嘌呤) 2.0 mg/L,分化率和平均分化芽数为81.35%和7.04个;不定芽增殖最佳培养基为MS+NAA(萘乙酸) 0.05 mg/L+6-BA 3.0 mg/L,增殖系数达到5.22;壮苗最佳培养基为MS+NAA 0.1 mg/L+6-BA 1.0 mg/L,平均苗高达到5.59 cm;不定芽生根最佳培养基为1/2 MS+NAA 0.2 mg/L,生根率和平均生根数最高,分别为82.54%和4.38条。本研究建立的三角梅下胚轴再生技术,为后续离体突变体诱导、多倍体育种及遗传转化体系的建立提供了技术保障。

Abstract:

To establish a stable and effective in vitro regeneration protocol, in the present investigation, the seed hypocotyl of Bougainvillea glabra‘Bouform 1'was used as the explant, and the growth status of induced differentiation, adventitious buds proliferation, strong seedling, rooting and so on was evaluated on MS medium supplemented with various plant growth regulators. The results showed that the optimal medium for adventitious differentiation was MS+IBA 0.1 mg/L+6-BA 2.0 mg/L, with the differentiation rate at 81.35% and the differentiated buds at 7.04 on average. The medium for adventitious buds proliferation was MS+NAA 0.05 mg/L+6-BA 3.0 mg/L, with the proliferation coefficient at 5.22. The seedling strength medium was MS+NAA 0.1 mg/L+6-BA 1.0 mg/L and the average seedling height reached 5.59 cm. The rooting medium was 1/2 MS+NAA 0.2 mg/L, with the rooting rate at 82.54% and 4.38 roots per seedling on average. The Bougainvillea hypocotyl regeneration technology established in this study provides technical support for subsequent in vitro mutant induction, polyploidy breeding and the establishment of genetic transformation systems.
