卷曲乳杆菌A7的益生特性及基因组测序分析

为评估一株源自健康女性阴道的卷曲乳杆菌(Lactobacillus crispatus)的益生潜力,本研究对其生长特性、生化功能、耐酸耐胆盐性能、产酸能力、药敏性和细胞粘附性进行系统分析,并与其他菌株进行全基因组系统发育分析,该菌株被鉴定为卷曲乳杆菌,将其命名为A7。结果表明,其生长迅速,能发酵乳糖和葡萄糖,具有良好的耐酸和产酸能力,但胆盐耐受性较弱。药敏实验显示,不同抗生素对其抑菌效果存在显著差异。粘附实验证实,该菌株能以浓度和时间依赖性的方式有效粘附于He La和C33a宫颈癌细胞。全基因组测定表明,其基因组全长为2 280 754 bp,共预测编码2 234个基因,总长度为2 004 369 bp,占基因组全长的87.88%,不存在质粒;通过基础数据库GO、COG、KEGG和CAZy注释,分别匹配到1 765个、1 816个、1 408个和78个基因的蛋白序列;通过CARD数据库中注释到111个耐药基因;基因组中还包括温度应激、氧化应激和细菌素合成等多种与益生功能相关的基因;共线性分析发现,同源卷曲乳杆菌之间整体基因组结构保守,但局部会呈现可辨别的结构变异。综上,该菌株具备良好的益生特性,尤其对宫颈癌细胞有显著粘附作用,有潜力开发为针对妇科健康的益生菌制剂。

Abstract:

This study aimed to evaluate the probiotic potential of a Lactobacillus crispatus strain isolated from the vagina of a healthy woman. A systematic analysis was conducted on its growth characteristics, biochemical functions, acid and bile salt tolerance, acid production capacity, antibiotic susceptibility, and performed phylogenetic analysis based on whole-genome-sequencing. The strain was identified as Lactobacillus crispatus and designated A7. The results indicated that it exhibits rapid growth and ferments both lactose and glucose. Furthermore, it demonstrated good acid tolerance and acid production capacity, but its tolerance to bile salts was relatively weak. Antibiotic susceptibility testing revealed significant differences among various antibiotics. Adhesion assays confirmed that the strain could effectively adhere to He La and C33a cervical cancer cells in a concentration-and time-dependent manner. Whole-genome sequencing determined that the complete genome of Lactobacillus crispatus A7 comprises 2 280 754 bp, encoding 2 234 predicted genes spanning 2 004 369 bp, which corresponds to 87. 88% of the total genome length. No plasmid sequences were identified. Genome annotation identified 1 765, 1 816, 1 408 and 78 genes assigned to GO, COG, KEGG and CAZy databases, respectively. The CARD database annotated 111 antibiotic resistance genes. The genome also contained various probiotic-related genes involved in temperature stress, oxidative stress, and bacteriocin synthesis. Synteny analysis revealed that the overall genomic structure of homologous L. crispatus strains is conserved, though local distinguishable structural variations were observed. In conclusion, this strain possesses promising probiotic properties, notably a significant adhesion capability to cervical cancer cells, suggesting its potential for development into a probiotic formulation for gynecological health.
