过度捕捉及贸易已严重威胁许多鸟类。然而,准确的物种鉴定是开展保护工作的首要前提。暗绿绣眼鸟(Zosterops japonicas)和灰腹绣眼鸟(Z. palpebrosa)都是国内深受喜爱而被广泛捕捉和笼养的种类,它们形态识别的主要特征为胸腹中线的黄色带。本实验以41只国内分布的暗绿和灰腹绣眼鸟的ND2基因片断构建NJ树,结果呈现出Kimura-2P距离高达0.076的两个分支,但与依据有无黄色带而鉴定的物种不一致;合并GenBank上这两种绣眼鸟的ND2序列后构建的NJ树呈现多个分支,但实验个体只聚集在由暗绿绣眼鸟和由灰腹绣眼鸟西南亚种组成的两个高支持率的分支上。以贝叶斯和最大似然法构建的系统树也呈现同样的结果。这说明胸腹中线是否具有黄色带并不能准确地识别这两种绣眼鸟。比对照片发现灰腹绣眼鸟西南亚种的虹膜为白色,而暗绿绣眼鸟的为褐色,该特征可作为国内这两种绣眼鸟的物种识别依据,为开展保护工作提供一定帮助。
Abstract:Over-exploitation of wild bird has been a major threat worldwide. However, accurate species identification is a first step for conservation actions. Both the oriental white-eye(Zosterops palpebrosa) and the Japanese white-eye(Zosterops japonicas) were the popular pet birds that were widely hunted and traded. The two species were identified by a yellow mesial belly stripe according to references widely used in China. We collected 41 individuals of the two species in China and amplified partial sequence of ND2. NJ analysis indicated two highly diverged clades with kimura-2P distance 0.076, which were inconsistent with the species identified with the yellow mesial belly stripe. Analysis included all the Genbank available ND2 sequences showed that individuals we collected in this study were clustered with two highly supported lineages: One were composed of the Japanese white-eye and the other were the subspecies siamensis of the oriental white-eye. Phylogenetic trees based on Bayesian and Maximum likelihood inferences showed the same structures. Such finding indicated that the yellow mesial belly stripe was an insufficient indicator for the identification of the two species. After checked the photos we collected, we discovered that the Oriental white-eye has white iris and the Japanese white-eye brown. This difference could be used to identify the two species in China, which was expected to provide positive support for the conservation of the two species.
