为了考察餐后运动时间对Ⅱ型糖尿病能量和脂代谢的影响,本研究通过高脂饮食和尾静脉注射60 mg/kg的链脲佐菌素来建立Ⅱ型糖尿病大鼠模型,大鼠分别在餐后0.5 h和餐后1 h进行30 min (15 m/min)的跑台运动,共运动4周。运动完成后,分别检测了各组大鼠能量代谢指标(Glu, Lac, BUN和NEFA)、血脂代谢指标(TG, TC, LDL-C和HDL-C)、糖化血红蛋白(HbA1c)、胰岛素(INS)和瘦素(Lep)水平。通过Western blotting检测大鼠胰腺组织中葡萄糖转运蛋白(GLUT-4)和胰岛素受体底物-1 (IRS-1)的蛋白表达。研究显示,与餐后0.5 h运动组相比,餐后1 h运动组的Glu和BUN水平显著降低。与餐后0.5 h运动组相比,餐后1 h运动组的HbA1c、INS和Lep水平显著降低。与餐后0.5 h运动组相比,餐后1 h运动组的TG、TC和LDL-C水平均显著降低,而HDL-C水平显著升高。与餐后0.5 h运动组相比,餐后1 h运动组的GLUT-4和IRS-1蛋白表达水平均显著升高。总之,餐后运动可有效改善Ⅱ型糖尿病的能量代谢和脂代谢,提高胰岛素敏感性,减弱胰岛组织损伤。餐后1 h运动是Ⅱ型糖尿病患者的理想运动方案。
Abstract:To investigate the effect of postprandial exercise time on energy and lipid metabolism in type Ⅱ diabetes,in this study, a type Ⅱ diabetic rat model was established by a high-fat diet and tail vein injection of 60 mg/kg of streptozotocin. Rats performed treadmill exercise for 30 min(15 m/min) at 0.5 h and 1 h after meals for 4 weeks.After exercise, rats in each group were tested for energy metabolism indicators(Glu, Lac, BUN, and NEFA), blood lipid metabolism indicators(TG, TC, LDL-C, and HDL-C), glycated hemoglobin(HbA1 c), insulin(INS), leptin(Lep) levels. Western blotting was used to detect the protein expression of glucose transporter(GLUT-4) and insulin receptor substrate-1(IRS-1) in rat pancreatic tissue. Studies showed that compared to the 0.5 h post-meal exercise group, the Glu and BUN levels in the 1 h post-meal exercise group were significantly reduced. Compared with the0.5 h post-meal exercise group, the HbA1 c, INS, and Lep levels in the 1 h post-meal exercise group were significantly reduced. Compared with the 0.5 h post-meal exercise group, the TG, TC, and LDL-C levels of the 1 h post-meal exercise group were significantly reduced, while HDL-C levels were significantly increased. Compared with the 0.5 h post-meal exercise group, the expression levels of GLUT-4 and IRS-1 protein in the 1 h post-meal exercise group were significantly increased. In short, postprandial exercise can effectively improve the energy and lipid metabolism of type Ⅱ diabetes, increase insulin sensitivity, and reduce islet tissue damage. 1 h exercise after meals is an ideal exercise program for patients with type Ⅱ diabetes.
