hsa-miR-375和hsa-miR-183-5p在子宫内膜癌中的表达及意义

摘要:

为探究hsa-miR-375和hsa-miR-183-5p在子宫内膜癌中的表达及意义,本院收集了子宫内膜癌患者手术治疗后切下的组织样本,应用实时荧光定量PCR法检测hsa-miR-375和hsa-miR-183-5p的表达;构建hsa-miR-375和hsa-miR-183-5p过表达质粒,转染细胞后,应用细胞锚定非依赖生长分析法检测细胞的增殖能力,Annexin V-FITC/PI流式检测细胞的凋亡比例,实时荧光定量PCR法检测凋亡相关基因m RNA的表达。结果显示,人子宫内膜癌组织和细胞系HEC-1B中,hsa-miR-375和hsa-miR-183-5p表达显著降低(p<0.05);hsa-miR-375和hsa-miR-183-5p过表达质粒转染HEC-1B后,细胞增殖能力明显降低,细胞凋亡百分比提高,TP53、Bax的mRNA表达水平显著提高,Bcl-2的mRNA表达水平显著降低(p<0.05)。研究结果表明,hsa-miR-375和hsa-miR-183-5p可通过调控凋亡相关基因的表达促进HEC-1B细胞的凋亡,抑制HEC-1B细胞的增殖,表现出抗肿瘤活性,在子宫内膜癌中具有潜在的应用价值。

Abstract:

To investigate the expression of hsa-miR-375 and hsa-miR-183-5 p in endometrial carcinoma and its clinical significance. We excised tissue samples after surgical treatment of endometrial cancer patients were collected, and the hsa-miR-375 and hsa-miR-183-5 p expression levels were measured by qRT-PCR. Establish the overexpression plasmid of hsa-miR-375 and hsa-miR-183-5 p. The endometrial cancer cells were transfested with overexpression plasmid and the proliferation ability of cells were detected by anchorage-independent growth assay.The proportion of apoptotic cells were measured by flow cytometry after Annexin V-FITC/PI staining. The apoptosis-related gene mRNA levels were measured by qRT-PCR. The expression level of hsa-miR-375 and hsa-miR-183-5 p was significantly decreased in human endometrial carcinoma tissues and cell line HEC-1 B(p<0.05).After the overexpression plasmid transfecting the HEC-1 B cells, the cell proliferation ability was significantly reduced, and the proportion of apoptotic cells was increased. The m RNA expression levels of TP53 and Bax was significantly increased, while the expression levels of Bcl-2 was significantly decreased(p<0.05). The hsa-miR-375 and hsa-miR-183-5 p can promote the apoptosis and inhibit the proliferation of HEC-1 B cells and exert its anticancer effects, by regulating the expression of apoptosis-related genes, showing its potential clinical applications in endometrial cancer.
