蛋白质组学作为后基因组时代新兴起来的交叉学科,已成为21世纪生命科学研究的前沿和热点。双向电泳(two-dimensional electrophoresis,2-DE)凭借其简便、快捷、高分辨率、高灵敏度,以及能将上万种蛋白质同时分离、分析等优点,已成为蛋白质组学研究的核心技术,在分子植物病理学研究中发挥着不可替代的作用。为此,本研究对蛋白质组学的发展、2-DE技术的基本原理、特点以及在分子植物病理学研究中的应用进行了阐述,并对其未来的发展方向作了展望。
Abstract:Proteomics, as an emerging interdiscipline in the post-genome era, has been the foreland and hotspot in study of life science in the 21 stcentury. Two-dimensional electrophoresis(2- DE) technology, which has the advantages of convenience, shortcut, high resolution, high sensitivity and simultaneously resolving tens of thousands of proteins in one separation and analysis procedure, as a core technology of proteomics, is playing an irreplaceably important role in research of molecular plant pathology. Hereby, this paper reviews the development of proteomics, the basic principles and characteristics of 2- DE technology, and its application in study of molecular plant pathology. In addition, the authors also look forward to the development direction of 2- DE technology for the future.
