响应面法优化色氨酸合成酶合成2-L-甲基色氨酸

本试验研究了色氨酸合成酶合成2-L-甲基色氨酸,验证色氨酸合成酶基因工程菌活性,用单因素实验及响应面分析法对色氨酸合成酶合成2-L-甲基色氨酸的合成条件进行优化。结果表明,最佳的合成条件:pH=8.06,温度为33.3℃,L-丝氨酸浓度为12.7 g/L,此条件下,手性药物2-L-甲基色氨酸的合成量为10.5 g/L。研究结果可为手性药物2-L-甲基色氨酸工业化生产提供参考价值。

Abstract:

Synthesis of 2-L-methyl t ryptophan by tryptophan synthase was studied and activity of tryptophan synthetase gene engineering bacteria was verified in this experiment. The synthesis reaction conditions of2-L-methyl tryptophan by tryptophan synthase were optimized with single-factor experiment and response surface methodology. The results showed that the optimal synthesis conditions were that: pH was 8.06, temperature was 33.3℃, concentration of L-serine was 12.7 g/L. Under this condition, synthesis quantity of chiral drug2-L-methyltryptophan was 10.5 g/L. This research should provide a reference for industrial production of chiral drug 2-L-methyltryptophan.
