水稻细菌性条斑病菌(Xamthomonas.oryzae pv.oryzicola,Xoc)是水稻的主要病原菌之一,其引发的水稻细菌性条斑病可造成水稻严重减产。本文利用Xoc GX01作为实验菌株,通过细菌培养、蛋白提取、纯化获得总蛋白,利用高p H值C18反相色谱作为第一维分离方式,再将收集的馏分通过LC-MS/MS技术得到了总蛋白质表达谱,共鉴定蛋白质1 621个,同时利用生物信息学技术对鉴定蛋白进行基因本体(gene ontology,GO)生物学分类,对生物功能、生物进程以及细胞组件进行了注释。为明确植物病原菌的作用机理和研究植物-病原微生物互作提供了方法学基础。
Abstract:Xamthomonas. oryzae pv. oryzicola(Xoc), the causal agent of rice bacterial leaf streak, is one of the most serious diseases of rice. It can cause a severe reduction of rice output. In this study, Xoc GX01 isolated was used as experimental strain, we got the total proteins by bacterial culture, extraction and purification of protein. Firstly,used high p H C18 reverse phase chromatography as the first dimensional separation, then got the intracellular protein expression spectrum using LC-MS/MS, finally identificated 1 621 proteins. Meanwhile biological function,biological process and cellular component had been carried on the Gene Ontology(GO) annotation. This result provides the basis for the further study of the mechanism of the plant pathogenic bacteria and plant-pathogenic microorganism interactions of Xanthomonas oryzae pv. oryzicola.
