肺癌H460细胞顺铂耐药相关基因的生物信息学筛选及预后价值分析

摘要:

利用生物信息学方法筛选肺癌H460顺铂耐药细胞(H460/DDP)中的差异表达基因,从Gene Expression Omnibus(GEO)公共数据库获得肺癌H460及其顺铂耐药细胞株的基因表达芯片GSE21656,利用GEO2R软件分析差异表达基因,利用DAVID在线数据库对差异基因进行重注释和功能富集分析。本研究共筛选出相关的差异表达基因75个,与H460细胞相比,H460/DDP细胞中有24个表达上调,51个表达下调,差异均具有统计学意义。预后分析的结果表明,高表达RAB3C[HR 1.33(1.131.56),p<0.01]、SERPINB1[HR 1.32(1.121.56),p<0.01]、CHRNA9[HR 1.25(1.11.42),p<0.01]、FAM83A[HR 1.27(1.081.5),p<0.01]、IGFBP4[HR 1.35(1.191.53),p<0.01]和IGF2BP1[HR 1.47(1.241.73),p<0.01]基因的肺癌患者拥有较差的总生存期。

Abstract:

In order to identify the cisplatin resistance related genes of lung carcinoma H460 cell by bioinformatics analysis.The m RNA microarray dataset GSE21656 was downloaded from the Gene Expression Omnibus database and analyzed by GEO2 R to obtain differentially expressed gene probes.Re-annotate and functional enrichment analysis of the probes were performed for differentially expressed genes using DAVID database.It was showed that a total of 75 differentially genes were identified from GSE24876,which including 24 up-regulated genes and 51 down-regulated genes in cisplatin resistance H460 cell(H460/DDP) compared with cisplatin sensitive H460 cell.Among them,the high expression of RAB3 C [HR 1.33(1.131.56),p<0.01],SERPINB1 [HR 1.32(1.121.56),p<0.01],CHRNA9 [HR 1.25(1.11.42),p<0.01],FAM83 A [HR 1.27(1.081.5),p<0.01],IGFBP4 [HR 1.35(1.191.53),p<0.01]and IGF2 BP1 [HR 1.47(1.241.73),p<0.01]were associated with worse overall survival for lung cancer patients.
