白花蛇舌草对人胃癌SGC-7901细胞增殖的影响

本研究以不同浓度的白花蛇舌草注射液处理人胃癌SGC-7901细胞,探究其对细胞增殖的影响。利用台盼蓝拒染法和吉姆萨染色法分别检测不同浓度的白花蛇舌草注射液对人胃癌SGC-7901细胞存活率和细胞克隆形成率的影响;采用EdU法检测药物处理后SGC-7901的细胞增殖率;最后应用RT-qPCR和Western blot分别检测药物处理后对细胞周期相关蛋白CCNB1和CCND1在mRNA水平和蛋白水平表达的影响。实验结果表明:白花蛇舌草注射液可抑制人胃癌细胞SGC-7901的细胞活力,其细胞存活率随着药物浓度的升高而降低;不同浓度的白花蛇舌草注射液处理人胃癌SGC-7901细胞48 h后,其细胞克隆形成率和细胞增殖率显著降低;RT-qPCR和Western blot结果显示,CCNB1和CCND1基因的mRNA及蛋白表达水平随着药物浓度的升高而显著降低。综上所述,白花蛇舌草通过下调CCNB1和CCND1基因的表达抑制人胃癌SGC-7901细胞增殖。

Abstract:

In this study, gastric cancer SGC-7901 cells were treated with Hedyotis diffusa injection in different concentrations in order to detect the effect on cell proliferation. Trypan blue and giemsa staining method were used to detect the effects of different concentrations of Hedyotis diffusa on SGC-7901 cell viability and cell clone formation. The cell proliferation rate of SGC-7901 after drug treatment was detected by EdU method.RT-qPCR and Western blot were also performed to determine the mRNA and protein levels of CCNB1 and CCND1. The results showed that Hedyotis difflosa injection could inhibit the cell viability of SGC-7901, and the cell viability decreased with the increase of drug concentration. Human gastric cancer cells SGC-7901 were treated with different concentrations of Hedyotis diffusura injection for 48 h, and the cell clone formation rate and cell proliferation rate were sig-nificantly decreased. RT-qPCR and Western blot analysis showed that the mRNA and protein levels of CCNB1 and CCND1 were significantly downregulated by Hedyotis diffusa injection in different concentrations. With the higher drug concentration, the expressions of CCNB1 and CCND1 showed a declining trend. In conclusion, Hedyotis diffusa suppresses the proliferation of gastric cancer SGC-7901 cells by inhibiting the expression of CCNB1 and CCND1.
