本研究采用DNA混合池扩增并进行直接测序的方法,对3个引进猪种和3个地方猪种Nramp1基因外显子2的SNP位点进行筛选,并通过生物信息学软件对该基因的m RNA二级结构、蛋白质二级结构和功能进行预测。结果在2个引进猪种(大白猪,长白猪)和3个地方猪种(八眉猪,蕨麻猪,烟台黑猪)中共检测到2个SNP位点,分别为T62C和A92G,而在杜洛克猪中未检测到SNP位点。生物信息学分析结果表明,T62C和A92G位点均降低了RNA二级结构的稳定性,Nramp1基因可能在转录和信号转导过程中发挥着重要作用。本研究结果丰富了猪Nramp1基因库,为进一步研究Nramp1基因功能和筛选抗病育种相关SNP位点提供理论参考。
Abstract：The single nucleotide polymorphism(SNP) sites of Nramp1 exon2 in three introduced and three local pig breeds were screened by DNA mixed pool amplification and direct sequencing,and the m RNA secondary structure,protein secondary structures and protein functions were predicted through the bioinformatics software.The results showed 2 SNP sites(T62C and A92G) were detected in two introduced pigs(Large White,Landrace) and three local pigs(Bamei,Juema,Yantai Black),and no SNP site was detected in Duroc pigs.The results of bioinformatics showed T62C and A92G sites both reduced the stability of secondary structure of RNA,and Nramp1 gene might play an important role in transcription and signal transduction.The study could enrich the Genbank of Nramp1 and supply theoretical references for further functional research of Nramp1 gene and screening of SNP sites related to disease resistance breeding.
