为了探讨补青颗粒治疗db/db小鼠(Mus musculus)糖尿病性白内障(diabetic cataract, DC)的效果及其作用机制,本研究特选取了8只db/m小鼠作为空白组,糖尿病模型鼠db/db小鼠40只,并随机分为模型组、补青颗粒高中低剂量组、杞菊地黄丸组,每组8只。空白组和模型组采用等量生理盐水灌胃,其余四组分别采用补青颗粒(高剂量4 mg/L,中剂量2 mg/L,低剂量1 mg/L)和杞菊地黄丸(1 mg/L)混悬液进行灌胃,干预时间为10周。测量小鼠体重,结果显示补青颗粒和杞菊地黄丸在一定程度上减轻了小鼠的体重;分离db/db小鼠晶状体并观察晶状体的混浊程度,发现补青颗粒和杞菊地黄丸组的晶状体清晰程度均明显优于模型组;用HE染色评估小鼠晶状体的病理损伤,发现补青颗粒组和杞菊地黄丸组的晶状体病理损伤程度均轻于模型组;通过免疫组化的方法检测Beclin1、Bcl-2、m TOR基因在小鼠晶状体中的表达,发现与模型组相比,补青颗粒组Beclin1蛋白表达量增加,mTOR、Bcl-2蛋白表达量减少。补青颗粒通过减轻db/db小鼠体重、晶状体浑浊度和病理损害,增加晶状体内的细胞自噬水平,从而对小鼠的早中期DC起到治疗作用。
Abstract:In order to investigate the effect and mechanism of Buqing granule on diabetic cataract(DC) in db/db mice(Mus musculus), 8 db/m mice were selected as the blank group, and 40 db/db diabetic mice were randomly divided into model group, Buqing granule high, medium and low dose group, Qiju Dihuang Pill group, with 8 mice in each group. The blank group and model group were gavaged with equivalent normal saline, and the other four groups were administrated with Buqing Granule(high dose 4 mg/L, medium dose 2 mg/L, low dose 1 mg/L) and Qiju Dihuang Pill(1 mg/L) suspension respectively. Each group was intragastric administation for 10 weeks. The weight of the mice was measured and the results showed that both Buqing Granule and Qiju Dihuang Pill reduced the weight to some extent. With the lens of db/db mice separated, the turbidity of lens was observed. It was found that the clarity of lens in Buqing Granule group and the Qiju Dihuang Pill group were significantly better than that in the model group.HE staining was used to evaluate the pathological damage of the mouse lens. The results showed that the pathologic lesion of the lens in the Buqing Granule group and the Qiju Dihuang Pill group were lower than that in the model group. The expression of Beclin1, Bcl-2 and m TOR genes in lens were detected by immunohistochemical method, and the results indicated that the expression of Beclin1 gene increased in Buqing granule group, while the expression of mTOR and Bcl-2 gene decreased in Buqing granule group compared with model group. Buqing granule could reduce weight, turbidity of lens and pathologic lesion of db/db mice, and increase the level of autophagy in the lens. Thereby, Buqing granule might have a therapeutic effect on the early and middle stage of DC in mice.
