摘要:

分选蛋白家族是生物进化过程中相对保守的一类蛋白,其结构上含吞噬细胞氧化酶同源性结构域(PX domain)结构域,与膜结构中的磷酸肌醇发生紧密结合,在内含体介导的分选运输、再循环利用和信号转导中起着重要作用。本研究主要从细胞内逆向运输网络分选途径梳理分选蛋白家族的调控作用,及其与逆运复合体retromer共同参与的内含体分选途径;并对现有的分选蛋白家族成员依据所含结构域的不同进行分类;同时,探求分选蛋白家族与相关疾病的内在联系,及其在疾病防治中作为潜在治疗靶点的可能性。

Abstract:

Sorting nexins family is a relatively conserved class of proteins in the evolutionary process of organisms.Its structure contains Phox domain,which is closely bound to phosphatidylinositol in the endosomal membrane.SNXs are involved in the process of retromer-mediated sorting pathways,cargo retrieved and signal transduction.In this review,we summarized SNXs classification,construction and mechanism of endosomal sorting.Moreover,we revealed the inner connection of SNXs with related diseases,and its potential as a potential therapeutic target in disease prevention and treatment.
