为了探索沉默MDR1基因后阿霉素对胃癌耐药细胞株SGC7901/ADM细胞凋亡的影响,将RNA干扰载体p-2.1-3转染至SGC7901/ADM细胞,并靶向沉默MDR1基因,命名SGC7901/ADM-2.1-3细胞。将转染干扰片段的SGC7901/ADM-2.1-3细胞及未转染的耐药细胞株SGC7901/ADM细胞和胃癌细胞SGC7901均用阿霉素处理48 h,MTT法检测细胞存活率,激光共聚焦显微镜下观察细胞形态,琼脂糖凝胶电泳检测DNA断裂情况。实验结果表明转染后SGC7901/ADM细胞IC50值和耐药指数均显著降低,SGC7901/ADM-2.1-3细胞相对逆转率达(80.51±0.989),激光共聚焦显微镜观察,细胞呈现明显的边缘化及脱落等凋亡特征,DNA琼脂糖凝胶电泳出现梯形断裂条带,呈现凋亡的特征。本研究证明,将MDR1基因沉默能够逆转胃癌耐药细胞株SGC7901/ADM细胞对阿霉素的耐药性,转染干扰片段的SGC7901/ADM-2.1-3细胞对阿霉素的敏感性显著提高,促进耐药细胞株SGC7901/ADM细胞凋亡。
Abstract:To explore the effect of doxorubicin on apoptosis of gastric cancer cell line SGC7901/ADM after silencing MDR1 gene,the RNA interference vector p-2.1-3 was transfected into SGC7901/ADM cells and targeted to silence MDR1 gene,named SGC7901/ADM-2.1-3 cells.SGC7901/ADM-2.1-3 cells,untransfected drug-resistant cell line SGC7901/ADM cells and gastric cancer cell SGC7901 were treated with doxorubicin for 48 h.Cell viability was measured by MTT assay.Cell morphology was observed under laser confocal microscopy.And DNA fragmentation was detected by agarose gel electrophoresis.The results showed that the IC50 value and drug resistance index of SGC7901/ADM cells were significantly decreased after irradiation.The relative reversal rate of SGC7901/ADM-2.1-3 cells reached(80.51±0.989).By laser confocal microscopy,the cells showed obvious apoptotic features such asmarginalization and shedding.DNA agarose gel electrophoresis showed trapezoidal rupture bands,which showed apoptosis.This study demonstrates that silencing MDR1 gene can reverse the resistance of gastric cancer drug-resistant cell line SGC7901/ADM to doxorubicin.The sensitivity of SGC7901/ADM-2.1-3 cells which transfected with interference fragment to doxorubicin was significantly improved,and their apoptosis was promoted.
