野油菜黄单胞菌(Xanthomonas campestris pv. campestris, Xcc)能产生胞外多糖(exopolysaccharides,EPS),也称黄原胶(Xanthan gum)。黄原胶具有优良的流变学性质,广泛应用于食品、药品、化妆品和石油开采等行业。黄原胶分为两种,工业级和食品级。工业级黄原胶颜色较深,纯度较低;食品级黄原胶为白色,其处理过程复杂,成本较高。工业上鲜有尝试改造工程菌株提高黄原胶产量和品质。本研究从Xcc群体感应系统和菌黄素生物合成途径入手,改造其代谢过程,得到了1株高产"白原胶"的工程菌株ΔrpfBΔxanK。相比野生型,ΔrpfBΔxanK不仅合成黄原胶能力显著提高,而且合成的菌黄素含量显著降低。在5 L发酵罐体系培养发现ΔrpfBΔxanK发酵液呈白色,黄原胶产量高达27.44 g/L。上述结果为工业上提高黄原胶生产能力提供了新方法。
Abstract:Xanthan gum is a kind of exopolysaccharides produced by Xanthomonas campestris pv. campestris(Xcc).Because of its rheological properties, xanthan gum has been applied in many industries including food, pharmaceutical, cosmetic and oil drilling industries. Xanthan gum can be divided into two types, the industrial grade xanthan gum and the food grade one. Industrial grade xanthan gum has a deeper color with a lower purity, while food grade one possesses a white color, which is processed with a high cost. Little progress has been made regarding improving the yield of xanthan gum via genetically modifying the metabolic network of Xcc in industries. In this study, we successfully constructed a white xanthan gum high-producing strain called ΔrpfBΔxanK by modifying its DSF-mediated quorum sensing system and the biosynthesis of xanthomonadin. Compared to wild-type, ΔrpfBΔxanK is able to produce significantly more xanthan gum and less xanthomonadin pigment. In 5 L batch fermentation,the yield of white xanthan gum produced by ΔrpfBΔxanK can reach as high as 27.44 g/L. These results provide us with a new method of improving the yield of xanthan gum industrially.
