秋石斛侧芽外植体消毒方法的研究

摘要:

以秋石斛栽培品种‘088’新生侧芽为外植体,研究了2%次氯酸钠(2%NaClO)和0.1%升汞(0.1%HgCl2)单独消毒和组合消毒对秋石斛侧芽茎尖、中部茎段和基部茎段的消毒效果,探索适合秋石斛侧芽消毒的有效方法。实验结果表明:(1)0.1%HgCl2对秋石斛侧芽消毒污染率低、存活率高,效果比2%NaClO好,2%NaClO+0.1%HgCl2组合消毒效果优于单独使用一种消毒剂消毒;(2)秋石斛侧芽茎尖、中部茎段和基部茎段的最佳消毒方式分别为:2%NaClO浸泡10 min+0.1%HgCl2浸泡6 min、2%NaClO浸泡10 min+0.1%HgCl2浸泡8 min和2%NaClO浸泡10min+0.1%HgCl2浸泡10min。

Abstract:

Used new lateral buds of Dendriobium hybrida cultivar ‘088'as explants, the disinfection effect of2% NaClO and 0.1% HgCl2alone and in combination disinfection for stem tip, middle stem and basal stem of Dendriobium hybrida lateral buds was analyzed, and to explored the effective explants disinfection method for Dendriobium hybrida lateral buds. The research result showed that:(1) 0.1% HgCl2had better explants disinfection effect for Dendriobium hybrida lateral buds than 2% NaClO, and the in combination disinfection effect was better than alone disinfection.(2) The best disinfection method for stem tip, middle stem and basal stem of Dendrobium hybrida lateral buds was 2% NaClO immersed 10 min +0.1% HgCl2immersed 6 min, 2% NaClO immersed 10 min+0.1% HgCl2immersed 8 min and 2% NaClO immersed 10 min+0.1% HgCl2immersed 10 min,respectively.
