冠突曲霉是茯砖茶发花阶段的主要微生物,其在Na Cl作为渗透压调节下分别进入有性产孢和无性产孢两个不同的世代,为了考查钙信号途径在其特殊产孢过程中的作用,通过克隆冠突曲霉钙信号调控的关键基因钙调素cam基因,并对这一基因进行结构和表达分析。结果表示:冠突曲霉中的cam基因全长924 bp,开放阅读框为450 bp,含6个内含子,预测编码149个氨基酸。cam参与了冠突曲霉的产孢及盐压力响应过程,cam基因对其无性发育产生正调控。实验结果为钙信号调控丝状真菌盐压力应答下的产孢机制研究奠定基础。
Abstract:The Aspergillus cristatus was the dominant fungus in the Fuzhuan tea,A.cristatus possess the asexual development stage in 3 mol/L Na Cl and sexual development stage in 0.5 mol/L Na Cl.In order to explore whether the Ca2+signaling pathway responds to the changes in the salt stress in A.cristatus,cam gene of A.cristatus was cloned and characterized.The full length of cam gene c DNA of A.cristatus is 924 bp,including a 450 bp open reading frame encoding 149 amino acids.The expression levels of cam gene are up-regulated in the asexual stage.The study results provide the basis for further exploring the relationship between the salt stress response and spore germination mechanism in fungi.
